Durch die Wette mit dem Herrn in der Szene "Prolog im Himmel" will Mephisto versuchen, den Knecht des Herren, Doktor Faust, der die eigene Wissbegierde niemals stillen kann, seine "Straße sacht zu führen!" (Z. 314), d.h. ihn vom rechten Weg abzubringen und ins Nichts zu führen. Da Mephisto sich selbst vorstellt als einen „Geist, der stets verneint!“ und dessen „eigentliches Element“ „Zerstörung, kurz, das Böse“ ist, ist es schon vorbestimmt, dass sein Einfluss auf die Liebenden, Faust und Gretchen, negativ sein wird. Auch wenn es Mephistos Verdienst ist, dass die beiden zusammenkommen, weil Faust es nicht schafft, Gretchen alleine zu umwerben, und dass aus der Verliebtheit unglücklicherweise Liebe wird, wird Mephisto sie zerstören, denn das liegt in seiner Natur. Dies geschieht vor allem, weil Mephisto nicht im Stande ist zu begreifen, was das Wort Liebe bedeutet. Er kennt nur das Sinnliche an einer Beziehung zwischen Mann und Frau und so versucht er, Faust soweit zu bringen, dass er in der sinnlichen Liebe zu Grunde geht und dass er aufhört, dem Sinn des Lebens nachzuspüren.

Welche Rolle spielt Mephisto beim Zusammenkommen der Verliebten? Schon in der "Hexenküche" versucht Mephisto, Faust mit dem Verjüngungstrunk so zu verzaubern, dass dieser "mit diesem Trank im Leibe, / Bald Helenen in jedem Weibe" (Z. 2604f.) sieht. Es ist also anzunehmen, dass Faust sich nur in Gretchen verliebt, weil er verhext wurde.

Mephistos hat eine Strategie, um Gretchen für Faust zu erobern. Weil sich aber Faust in Gretchen verliebt und vor allem umgekehrt, gerät die Sache für Mephisto außer Kontrolle und er muss zum Gegenspieler Gretchens werden. Beide streiten um Faust und die Handlung erweitert sich zur Frage, ob es Mephisto gelingen wird, Faust von seiner Geliebten zu trennen und ihre Liebe zu zerstören.
